Your best bet is to take the AirTrain which takes you from Newark Airport to New York - Penn Station in roughly 30 minutes. So yes, you do have enough time to explore NYC for a little while. The ride may cost up to $20 - not sure.

Alternatively, you can take the PATH train from Newark-Penn Station to either World Trade Center, West Village (Christopher St.) or Chelsea (23 St.). However you will need to hop on the Amtrak from Newark to Newark Penn Station first. That ride is about 3 mins and you can probably get away with not paying for it. Cost for this option is $1.50 for the cost of a PATH ticket, but it will take about 1 hour if you have to transfer at Journal Square.

There is so much to do in NYC so it totally depends what you are into. You would have enough time to visit one of the following:

-the MOMA and have a nice brunch

-walk around Soho and shop

-go to the top of the Empire State Building

-explore lower Manhattan and see South St. Seaport

Have fun.
